Targeted Mass Spectrometric Immunoassay for Detection of Cystatin C Isoforms in Cerebrospinal Fluid

Abstract: Targeted cerebrospinal fluid (CSF) Mass Spectrometric Immunoassay for cystatin C was developed and applied to a small number of CSF and serum samples obtained from healthy and individuals diagnosed with multiple sclerosis. Cystatin C and its variants were retrieved from the CSF samples via affinity micro columns derivatized with cystatin C antibodies, and were eluted onto MALDI mass spectrometer targets. The resulting mass spectra revealed various levels of cystatin C isoforms in the samples. Truncated Cystati… Show more

“…In addition to CKD, CysC has been related to CVD and different neurological disorders. While higher levels of circulating CysC are associated with increased CVD risk, , increased expression and secretion in the brain may have both neurodegenerative as well as neuroprotective effects. , Proteoforms of CysC have been identified in blood samples and cerebrospinal fluids ,, and may give further insights into pathologies of CKD , and neurological disorders.…”

“…Moreover, paired serum/CSF samples origin from same individuals was screened. As presented in the previous studies, using the optimized MSIA qualitative assay, cysC and its wild type isoforms were detected in the analyzed CSF samples (Nedelkov, Shaik et al 2008). However, several correlations were observed when matched serum/CSF samples were analyzed.…”

“…Using immunoaffinity pipettes it was possible to extract only the targeted protein and to analyze it with MALDI-TOF-MS. Experimental conditions such as sample dilution and incubation time were optimized in order to provide better results (9).…”
