Targeted mechanical forces enhance the effects of tumor immunotherapy by regulating immune cells in the tumor microenvironment

Abstract: Mechanical forces in the tumor microenvironment (TME) are associated with tumor growth, proliferation, and drug resistance. Strong mechanical forces in tumors alter the metabolism and behavior of cancer cells, thus promoting tumor progression and metastasis. Mechanical signals are transformed into biochemical signals, which activate tumorigenic signaling pathways through mechanical transduction. Cancer immunotherapy has recently made exciting progress, ushering in a new era of “chemo-free” treatments. However,… Show more

“…The main ECM feature common to different tumor types is ECM stiffness, which highly impacts on immune cell infiltration representing a structural and physical barrier to the recruitment of effector T cells. An extreme matrix density and rigidity is also known to associate with impaired drug delivery to the tumors, thus pinpointing ECM stiffness as a double-edge sword deeply impaction on the efficacy of ICI ( 25 ).…”

“…Increased fibrillar collagen density and radial alignment of collagen are observed in invasive breast tumors and observed in physiologically normal breast involution and in PPBC ( 169 171 ). These ECM-based mechanisms inherently improve the motility and invasiveness of breast cancer cells, however, there are also well-documented interactions between ECM dynamics and the immune system, suggesting further downstream effects of the local mechanical involution environment ( 172 , 173 ). Beyond ECM dynamics, there is a distinct role for direct mechanical forces in regulating involution.…”

“…To further feed this fertile soil, cancer cells secrete soluble factors like chemokines, cytokines, and growth factors to actively recruit diverse cell types to the TME and reprogram them ( Mao et al, 2021 ). In turn, these cells in the TME facilitate the invasion and migration of cancer cells, their evasion from immune destruction, resistance to chemotherapy, and stimulation of angiogenesis, augmenting the tumor aggressiveness ( de Visser and Joyce, 2023 ; Zhu et al, 2023 ). Hence, a complete understanding of how cancer cells interact with their microenvironment is crucial to overcoming current challenges in cancer treatment.…”
