Temperature modulates virus‐induced transcriptional gene silencing via secondary small RNAs

Abstract: Virus-induced gene silencing (VIGS) can be harnessed to sequence-specifically degrade host transcripts and induce heritable epigenetic modifications referred to as virus-induced posttranscriptional gene silencing (ViPTGS) and virus-induced transcriptional gene silencing (ViTGS), respectively. Both ViPTGS and ViTGS enable manipulation of endogenous gene expression without the need for transgenesis.Although VIGS has been widely used in many plant species, it is not always uniform or highly efficient. The efficie… Show more

“…TaWRKY74 and TaCRR6 RNAi fragments were cloned into BSMV‐γ vectors for VIGS assays in T. aestivum (Yang et al., 2018) . NbWRKY17 and NbCRR6 RNAi fragments were cloned into TRV2 for VIGS in N. benthamiana (Fei et al., 2021). The primers and restriction enzymes used are listed in Table S1.…”

“…Lastly, the timing of the VIGS phenotype appearance and the duration for which the gene silencing effect lasts is speciesspecific. Also, an effect of the ambient temperature on VIGS through secondary siRNA production was reported (Fei, Pyott and Molnar, 2021).…”

“…As for sNRT2.3-1, the temperature shifts from 25° to 32°C changed the hairpin loop sequences from UGUU-GGAGAUGGAG into GAUGGA (Fig. 5, A and D), which would affect the binding to the target mRNA (14,15).…”
