Tetrahydroxystilbene glucoside ameliorates memory and movement functions, protects synapses and inhibits α-synuclein aggregation in hippocampus and striatum in aged mice

Abstract: TSG improved the memory and movement functions in aged mice through protecting synapses and inhibiting α-synuclein overexpression and aggregation in multiple brain regions. The results suggest that TSG may be beneficial to the treatment of ageing-related neurodegenerative diseases.

“…THSG has antioxidant and anti-inflammatory activity [14][15][16][17]. Previous studies have shown that THSG can promote neuronal growth factor expression and improve learning and memory function in rodents [18][19][20]. Furthermore, THSG can protect from oxidative stress-induced neuronal degeneration and neurotoxicity in vitro, and in animal models of Parkinsonʼs disease and Alzheimerʼs disease [21][22][23].…”

“…TSG also elicits neuroprotective effects in various neurodegenerative diseases and cerebral ischemia. In aged mice, TSG treatment rescues synapses and suppresses α -synuclein overexpression in the brain, subsequently improving their memory and movement functions [ 10 ]. TSG protects dopamine (DA) neurons against lipopolysaccharide- (LPS-) induced neurotoxicity through dual modulation on glial cells by attenuating microglial-mediated neuroinflammation and enhancing astroglial-derived neurotrophic effects [ 11 ].…”

“…In particular, macrophage plays an important role in the inflammation procession by inducing overexpression of a series of inflammatory cytokines and mediators [2, 5]. It has been reported that the extract of Polygonum multiflorum has various pharmacological activities, including antioxidant activities, anti-inflammation, delay senility, and improvement of memory ability [611]. As a major bioactive component of Polygonum multiflorum, TSG has been confirmed to inhibit the inflammatory processions in vivo and in vitro [12].…”
