During the recent decade, high-throughput sequencing (HTS) techniques, in particular, DNA metabarcoding, have facilitated increased detection of biodiversity, including harmful algal bloom (HAB) species. In this study, the presence of HAB species and their appearance patterns were investigated by employing molecular and light microscopy-based monitoring in Tokyo Bay, Japan. The potential co-appearance patterns between the HAB species, as well as with other eukaryotes and prokaryotes were investigated using correlation and association rule-based time-series analysis. In total, 40 unique HAB species were detected, including 12 toxin-producing HAB species previously not reported from the area. More than half of the HAB species were present throughout the sampling season (summer to autumn) and no structuring or succession patterns associated with the environmental conditions could be detected. Statistically significant (p < 0.05, rS ranging from −0.88 to 0.90) associations were found amongst the HAB species and other eukaryotic and prokaryotic species, including genera containing growth-limiting bacteria. However, significant correlations between species differed amongst the years, indicating that variability in environmental conditions between the years may have a stronger influence on the microalgal community structure and interspecies interactions than the variability during the sampling season. The association rule-based time-series analysis allowed the detection of a previously reported negative relationship between Synechococcus sp. and Skeletonema sp. in nature. Overall, the results support the applicability of metabarcoding and HTS-based microalgae monitoring, as it facilitates more precise species identification compared to light microscopy, as well as provides input for investigating potential interactions amongst different species/groups through simultaneous detection of multiple species/genera.

DIATOMS are unicellular microscopic eukaryotic algae that are found in both fresh and marine water. They are primary producers among phytoplanktons and responsible for fixing almost 25% of the atmospheric CO 2 (Ramachandra et al., 2009). Besides this, diatoms contribute to one fifth of the global biomass primary production (Leblanc et al., 2012). Unlike other algae, they have a unique feature of having cell wall being made up of silica which could tolerate harsh environmental conditions (Round et al., 1990). It is noteworthy that diatom morphology is distinct and determined mainly by the salinity, temperature, and other environmental factors of the water thus making their species diverse across fresh and marine waters (Trobajo et al., 2011). However, the leakage of marine diatoms to fresh waters has resulted into morphological transitions in diatoms (Alverson et al., 2007). Among diatom lineages, Theriot and Serieyssol (Theriot & Serieyssol, 1994) chose Thalassiosirales lineage to study the evolutionary transition between marine and fresh water diatoms. This is because Thalassiosira is essentially a marine species found mostly in marine water and rarely in fresh water. Among the completely sequenced diatoms, Thalassiosira pseudonana has a genomic history quite likely originated from a fresh water diatom (Alverson et al., 2011a;Armbrust et al., 2004). The phylogeny of Thallossiosira identified by chloroplast genes (psbC and rbcL) and nuclear genes (SSU and partial LSUrDNA) reveals that there are clades of marine and fresh water species of Cyclotella.

Harmful algal blooms (HABs) constitute a global problem, affecting aquatic ecosystems, human health, fisheries and local economies. The Bay of Bengal, along the Bangladesh coast, is exceedingly suffering from pollution or anthropogenic nutrification that influences frequently occurring HAB species. The progression of climate change and eutrophication invigorate HAB trends and responses that in turn affect the respective coastal livelihood and economic growth. Tripos spp., Dinophysis spp., Protoperidinium spp., Chaetoceros spp., and Pseudo-nitzschia spp. are the common bloom-forming HAB species in the coastal waters of Bangladesh. Despite having huge potentiality for regional and global perspectives, the coastal region of Bangladesh remains relatively unexplored compared to other regions in the context of HABs and their pernicious effects. As a result, harmful algal blooms and the accumulation of algal toxins may interrupt fisheries, aquaculture, aquatic ecosystems and public health in the country. Therefore, proper research on the biology and ecology of harmful algae, biotoxins and their relationship with environmental factors need to be adequately understood to minimize their adverse effects on the noted marine resources of the Bay. This review focused on an overview of the HAB related issues – causes of HABs, their occurrences and abundances, associated environmental factors and adverse effects in the coastal zone of Bangladesh.
