The 16S rRNA Analysis and Enzyme Screening of Bacillus from Rhizosphere Soil of Lombok Island

Abstract: Bacillus are commonly found in nature, especially in soil and food. It has the ability to produce bioactive compounds as well as the enzyme. This study was aimed to isolate, identify, and screen their enzyme activities. Four soil samples from Mandalika, Lombok Island, West Nusa Tenggara (NTB), were used for isolation. Bacillus was isolated using the heat-shock method and characterized through Gram staining, endospore staining, and morphological phenotype. Bacillus identification was conducted based on 16S rRNA… Show more

“…The plant material and coarse roots were removed from the soil and remaining soil was used for isolation of bacterial microbes. For isolation purposes, one gram soil sample was mixed in distilled sterilized water (10 mL) and incubated at 80 • C for 10 min in the hot water bath [27]. The whole mixture was serially diluted up to 10 −7 dilutions using distilled autoclaved water.…”

“…The results of DNA extraction were measured for purity using a nanodrop spectrophotometer (NanoDrop™ One/OneC). Furthermore, the amplification of the 16S rRNA gene was carried out using universal bacterial primers 27F (5'-AGAGTTTTGATCTGGTCCAG-3') and 1492R (5'-GGTTACCTTGTTACGACTT-3') (Sulistiyani et al 2021). The PCR mix used was consisted of 12.5 µL (2x) MyTaq HS Red Mix (BIO-25048), 9.5 µL doubledistilled water, 1 µL 10 µM 27F primer, 1 µL 10 µM 1492R primer, and 1 µL template DNA.…”
