The Alternatively Spliced Exon of the Platelet-Derived Growth Factor A Chain Encodes a Nuclear Targeting Signal

Abstract: We have previously shown that the SIS/platelet-derived growth factor B chain contains a nuclear targeting signal near its C terminus. Here we show that the platelet-derived growth factor A chain also contains a nuclear targeting signal encoded by an exon which is subject to alternative splicing. This sequence is capable of targeting a nonsecreted form of the A chain to the nucleus and can also target the cytoplasmic proteins dihydrofolate reductase, chloramphenicol acetyltransferase, and pyruvate kinase to the… Show more

“…After the first description of internalization of [ 3 H] labelled TRH to the nucleus (Gourdji et al, 1973) many others peptides have been detected in the nucleus of target cells (Rakowicz-Szulczynska et al, 1986;Smith and Jarett, 1987;Smith et al, 1988;Maher et al, 1989;Soler et al, 1989;Baldin et al, 1990;Morel, 1994;Wiedlocha et al, 1994Wiedlocha et al, , 1996Maher, 1996;Jans et al, 1997) including Growth hormone (GH) and its receptor (GHR). After its binding to the plasma membrane receptor, GH is internalized in different cellular compartments (Roupas and Herington, 1989;Ilondo et al, 1992;.…”

“…Maher et al (11) recently have shown that the nuclear targeting signal of the A chain of platelet-derived growth factor is dependent on alternative splicing. Thus, bFGF represents another example of an endogenous growth factor with two possible subcellular locations regulated at the posttranscriptional level.…”
