The Anti-Wrinkle Mechanism of Melatonin in UVB Treated HaCaT Keratinocytes and Hairless Mice via Inhibition of ROS and Sonic Hedgehog Mediated Inflammatory Proteins

Abstract: Though melatonin is known to improve ultraviolet B (UVB)-induced oxidative damage and inflammatory conditions via the blockade of the nuclear factor (NF)-κB, interleukin (IL)-6, there is no report on the anti-wrinkle effect of melatonin to date. Hence in the present study, the anti-wrinkle mechanism of melatonin was elucidated in UVB treated HaCaT keratinocytes and hairless mice. Herein melatonin protected against a radical initiator tert-Butyl hydroperoxide (t-BOOH) induced reactive oxygen species (ROS) produ… Show more

“…ROS induced by UVB irradiation contributes to intrinsic aging such as photoaging. In particular, ROS induce wrinkles by inducing degradation of the extracellular matrix (ECM) through induction of MMPs and elastase enzymes in keratinocytes and fibroblasts [32][33][34]. To confirm the antiwrinkle effect of AL, we measured its elastase inhibition activity [35].…”

“…ROS induced by UV irradiation mediate diverse cellular functions and alterations, including matrix metalloproteinases (MMPs) secretion and extracellular matrix (ECM) degradation [2]. It is well documented that prolonged UVB exposure induces skin aging, leading to wrinkle formation because of collagen and elastin fiber breakdown along with the inhibition of procollagen synthesis [3].…”

“…19,20 Melatonin increased the collagen content in cultured cells by activating membrane receptors of fibroblasts and myofibroblasts, 21 as well as increased procollagen in HaCaT keratinocytes. 22 In this research, we investigated the effects of different solvents for extraction of some by-products from purple waxy corn: cob and silk. The active compounds, the 3 anthocyanins (C3G, Pn3G, Pg3G) and melatonin, were determined.…”
