The antiarrhythmic peptide rotigaptide (ZP123) increases gap junction intercellular communication in cardiac myocytes and HeLa cells expressing connexin 43

Abstract: 1 We investigated the effects of rotigaptide (ZP123), a stable hexapeptide with antiarrhythmic properties, on gap junction mediated intercellular communication in contracting rat neonatal cardiac myocytes, HL-1 cells derived from cardiac atrium and in HeLa cells transfected with cDNA encoding Cx43-GFP, Cx32-GFP, Cx26-GFP, wild-type Cx43 or wild-type Cx26. 2 Intercellular communication was monitored before and after treatment with rotigaptide following microinjection of small fluorescent dyes (MWo1 kDa). The co… Show more

“…Rotigaptide augmented gap-junctional conductance and improved cell coupling (Shiroshita-Takeshita et al, 2007). Rotigaptide also increased coupling in cells expressing Cx43, but not those expressing Cx32 and Cx26 (Clarke et al, 2006).…”

“…It would be easy to determine the extent of myocardial salvage by GJC inhibition if there were a specific and direct opener of closed gap junctions. A new gap junction opener, rotigaptide (4,11,35), is apparently selective. However, it suppresses the dephosphorylation of Ser297 and Ser368 in Cx43 during ischemia, indicating the activation of a protein kinase and/or the inhibition of a PP.…”

“…27 However, another study failed to observe changes in the phosphorylation status of connexin 43 in response to rotigaptide. 28 Rotigaptide increases connexin 43 expression after 24 hours in neonatal rat cardiomyocytes, 29 but such an effect is unlikely to explain changes after short-term intravenous administration.…”
