The Anxiolytic Effect of Two Oriental Herbal Drugs in Japan Attributed to Honokiol from Magnolia Bark

Abstract: An improved elevated plus-maze test in mice revealed that seven daily treatments with two different traditional Chinese medicines, known as Kampo medicines in Japan, Hange-koboku-to (composed of extracts of 5 plants) and Saiboku-to (composed of extracts of 10 plants), produced an anxiolytic effect, and the effect was mainly due to the presence of honokiol derived from magnolia. This study was carried out to evaluate the anxiolytic potential of honokiol, Hange-koboku-to and Saiboku-to, which were prescribed wit… Show more

“…administration, its serum half-life is >5 h, and its plasma concentration is similar to the concentration needed to induce in vitro cytotoxicity against breast cancer (36)(37)(38)(39). Moreover, HNK administration was well tolerated and no weight loss or other major side effects were observed in this and in previous other animal studies (13,19,37). Thus, HNK administration, at effective doses to treat breast cancer, seems to be safe and feasible.…”

“…Both lignans increased ChAT activity and inhibited AChE activity in vitro, and increased hippocampal ACh release in vivo [78]. Honokiol and magnolol show anxiolytic effects [79] and these appear to be due to their ability to potentiate GABAergic neurotransmission [80]. These two compounds also appear to have antioxidant antiinflammatory and neuroprotective properties and such polyvalency in activity is of interest in their potential use in treating AD [81,82].…”

“…Du point de vue pharmacologique, Sb-to potentialise la baisse de libération d'acétylcholine induite par le diazépam. Les préparations Hk-to ou Sb-to, dont a retiré le magnolia, n'ont pas d'activité anxiolytique [13]. L'effet anxiolytique du honokiol est supérieur à celui du magnolol.…”
