Transcriptomic data can be used to predict environmentally impacted phenotypic traits. This type of prediction is particularly useful for monitoring difficult-to-measure phenotypic traits and has become increasingly popular for monitoring high-value agricultural crops and in precision medicine. Despite this increase in popularity, little research has been done on how many samples are required for these models to be accurate, and which normalization should be used. Here we create a massive RNA-seq dataset from publicly availableArabidopsis thalianadata with corresponding measurements for age and tissue type. We use this dataset to determine how many samples are required for accurate model prediction and which normalization method is required. We find that Median Ratios Normalization significantly increases performance when predicting age. We also find that in the case of our dataset, only a few hundred samples are required to predict tissue types, and only a few thousand samples are necessary to accurately predict age. Researchers should consider these results when choosing the number of samples in a transcriptomic experiment and during data-processing.Author SummaryLarge datasets have become ubiquitous in both research and industry, with thousands and sometimes millions of samples being collected for a single project. In biology a prominent new technology is RNA-seq, which can be used to measure the expression level of thousands of genes for a single sample. These measurements are used for a variety of downstream applications, including predicting phenotypic traits (i.e. height, disease, etc.). A number of experiments have attempted to use RNA-seq data to make phenotype predictions with varying success. This is partially due to the small sample size of their experiments. RNA-seq datasets are currently relatively small--only a dozen to a few hundred samples--due to the cost per sample. This is expected to change as the cost of sequencing decreases. In this paper we create a massive conglomerate RNA-seq dataset from publicly availableArabidopsis thalianaRNA-seq data. We use this dataset to determine how many samples are required to accurately predict plant age and tissue type using machine learning models. We also explore the best way to normalize large datasets. Our results show the potential of massive RNA-seq datasets, and can be used to inform experimental design for phenotype prediction.

Through an ethnographic and comparative approach to counseling and psychotherapy, this article highlights the unique psychological mechanisms and cosmological perceptions involved in the practice of “reading the heart” ( du xinshu) in Chinese divination, particularly in the fortune-telling system based on bazi (one’s birth date and time). It reveals how diviners engage in a culturally specific dialogue that strategically influences clients’ cognitive, emotional, and moral experiences, how the professional ethics of divination can influence the ways in which practitioners guide customers, positively or negatively. Chinese divination, which emphasizes balance and a holistic worldview harmonizing society and nature, also compels diviners to pay great attention to reaching and touching customers’ hearts from an interpersonal perspective rather than resorting to mysterious supernatural forces. The focus of analysis is on the distinctive verbal skills and language performance of diviners, with a particular reference to a divination pedagogy manual that teaches communication skills to professional diviners. I argue that diviners’ language performance is intertwined with their effort to win the clients’ hearts and convictions in order to enhance their own authority and legitimacy. The distinctiveness of the Chinese indigenous approach provides a rich contrast to Western psychological practices. This analysis offers new insights on the therapeutic potential of divination as a culturally attuned method of counseling.

The “Fasting of the Heart” (Xinzhai 心斋): Zhuangzi, Daoism and Its Transformative Healing Power in Contemporary Psychotherapy

This article investigates a form of psychotherapy rooted in the Daoist notion of xinzhai, “fasting of the heart” in the Zhuangzi. Xinzhai is described as a meditative practice that “empties” the heart ( xin 心) so that it becomes responsive to the world, but is not affected by it. Also known as the “mirror-heart,” this state is characteristic of the Daoist sage. The xinzhai therapy is practiced together with Jung’s Active Imagination and Bosnak’s Embodied Imagination. In this hybrid therapy, the therapist’s “empty heart” aids in developing a non-judgmental openness in the client, allowing free flow of vital energy ( qi 气) that may lead to the dissolution of object-subject boundaries and inner transformation ( wuhua 物化). The article critically assesses both the merits and pitfalls of such Daoism-based hybridized therapies. It argues that therapies based on ancient spiritual self-cultivation, and more generally, the psychologization of Daoism and the hybridized use of the heart ( xin) and the psyche in the context of a far-reaching pragmatic governmental approach to ancient wisdom may dilute the spiritual depths of Zhuangzi’s teachings.
