The benefits of adipocyte metabolism in bone health and regeneration

Abstract: Patients suffering from musculoskeletal diseases must cope with a diminished quality of life and an increased burden on medical expenses. The interaction of immune cells and mesenchymal stromal cells during bone regeneration is one of the key requirements for the restoration of skeletal integrity. While stromal cells of the osteo-chondral lineage support bone regeneration, an excessive accumulation of cells of the adipogenic lineage is thought to promote low-grade inflammation and impair bone regeneration. Inc… Show more

“…rBMAT, which also expands in the proximal tibia during aging and obesity, has been described to contain more saturated fatty acids (9). The fatty acid composition in the proximal part of the tibia of CR mice was therefore more similar to distal BMAT, which indicates that the type of fat that forms upon CR might be more related to cBMAT, which arises early in life and is thought to play a more physiological beneficial role compared to rBMAT (17,41). Our data further show that fatty acid desaturation might occur directly in the bone as the expression of stearoyl-Coadesaturase 1 (Scd1) is upregulated upon CR and the ratio of monounsaturated palmitoleic acid over the saturated palmitic acid is consistently elevated.…”

“…Adiponectin is an adipocytokine that is intensively expressed in human adipose tissue [68][69][70][71][72]. It is secreted almost exclusively by adipocytes, mainly in visceral adipose tissue and, to a lesser extent, in peripheral adipose tissue and bone marrow.…”

“…Adiponectin also exhibits anti-osteoporotic activity, promoting differentiation and mineralization of osteoblasts and inhibiting the secretion of OPG [100]. Adiponectin stimulates the expression of osteocalcin, which acts as a hormone regulating glucose metabolism and fat mass [72,73,87,[101][102][103][104][105][106]. The potential effect of adiponectin on osteoblasts and osteoclasts, and therefore on bone remodeling, may be mediated through the endocrine system and may influence lipid metabolism.…”

“…Among these, adiponectin is the most abundant and is secreted predominantly by the white adipose tissue. Notably, adiponectin has anti-inflammatory and anti-atherogenic effects, and plays an important role in lipid and glucose metabolism by increasing insulin sensitivity, promoting the oxidation of free fatty acids, decreasing de novo synthesis/accumulation of lipids and protecting hepatic cells from apoptosis (23,24). The effects of adiponectin on the liver are mediated by its receptors (AdipoR1, AdipoR2), interacting with the adaptor protein phosphotyrosine interaction (APPL1).…”
