The Bergen COPD microbiome study (MicroCOPD): rationale, design, and initial experiences

Abstract: BackgroundRecent methodological developments, in particular new sequencing methods for bacterial RNA/DNA, have shown that microorganisms reside in airways that do not suffer from acute infection and that respiratory microbiota might vary according to airways disease status. We aim to establish high-quality sampling methods for lower airways microbiota as well as describe the respiratory microbiome in subjects with and without chronic obstructive pulmonary disease (COPD) and to relate the microbiome to disease … Show more

“…The MicroCOPD study included COPD and asthma patients as well as subjects without lung or airways disease (‘controls’) 8. Participants were recruited from the Bergen COPD Cohort Study9 and the GenKOLS Study,10 in addition to volunteers from the outpatient clinic at the Department of Thoracic Medicine, Haukeland University Hospital and asthma patients from a local pulmonology clinic.…”

“…In the Bergen COPD microbiome study (MicroCOPD)8 we performed more than 300 research bronchoscopies in subjects with and without obstructive pulmonary disease. The current analysis investigates if research bronchoscopy is less safe in subjects with obstructive lung disease by evaluating complications and discomfort occurring immediately, and within a week after bronchoscopy.…”

“…The design of the entire MicroCOPD study has been published previously [ 28 ]. The current analysis includes 58 control subjects, 64 subjects with COPD and three subjects with asthma.…”

“…In preparation for the analyses of a large, ongoing COPD microbiome study [ 28 ], we sought to reduce contamination as well as assess the performance of different sampling techniques. In the current paper we present analyses to examine the degree of oropharyngeal influence on the airway microbiome applying protected bronchoscopic sampling techniques.…”

“…Understanding why and how these diseases develop and progress over time requires long-term follow-up. Many cohorts have been established worldwide, such as ECLIPSE, CanCOLD, Birmingham, MicroCOPD ( 24 27 ). Although advancements have been made regarding descriptions of the main characteristics of chronic airway diseases, much is still to be explored about the factors associated with changes in the microbiome and the relevance of these changes in host-specific pathogenesis of the disease ( 23 ).…”
