The CD28-B7 Family in Anti-Tumor Immunity: Emerging Concepts in Cancer Immunotherapy

Abstract: The interactions between B7 molecules and CD28-family receptors are crucial in the regulation of adaptive cellular immunity. In cancer, the aberrant expression of co-inhibitory B7 molecules has been attributed to reduced anti-tumor immunity and cancer immune evasion, prompting the development of cancer therapeutics that can restore T cell function. Murine tumor models have provided significant support for the targeting of multiple immune checkpoints involving CTLA-4, PD-1, ICOS, B7-H3 and B7-H4 during tumor gr… Show more

“…In addition, expression of PD-1 ligands PD-L1/B7-H1 and B7-H4 in macrophages inhibits antitumor immune responses (36). We confirmed expression of SKAP1 was not altered in the thymus of SKAP2 À/À mice and that the level of B7-H1 and B7-H4 in macrophages was not affected by the absence of SKAP2 (Supplementary Fig.…”

“…42 However, almost none of the relevant review articles include BTLA as a B7-H4 receptor. 10,[43][44][45][46][47][48][49][50] Our findings provide additional support for the role of BTLA as a B7-H4 receptor.…”

“…It is now clear that suppressive effects mediated by the upregulation of inhibitory B7 family members within tumor microenvironments are an important factor in the failure of cytolytic lymphocytes to control tumors (40,41). A central issue for cellular immunotherapies is therefore avoiding inhibition of adoptively transferred cells by these pathways.…”
