The Clinical Relevance of the miR-197/CKS1B/STAT3-mediated PD-L1 Network in Chemoresistant Non-small-cell Lung Cancer

Abstract: Programmed cell death ligand-1 (PD-L1) has recently gained considerable attention for its role in tumor immune escape. Here, we identify a miR-197/CKS1B/STAT3-mediated PD-L1 network in chemoresistant non-small-cell lung cancer (NSCLC), independent of immunoinhibitory signals. miR-197 is downregulated in platinum-resistant NSCLC specimens, resulting in the promotion of chemoresistance, tumorigenicity, and pulmonary metastasis in vitro and in vivo. Mechanistic investigations reveal that a miR-197-mediated CKS1B/… Show more

“…And CKS1B induced phosphorylation of SATA3, which was about tumorigenesis (Yu et al, 2014). Moreover, Bcl-2 (a typical antiapoptotic factor), c-Myc (an oncogenic transcription factor), and cyclin D1 (a cell-cycle-regulating protein) were upregulated with inhibition of miR-197, three of which were oncogenic genes and ordinary transcriptional target genes of STAT3 (Fujita et al, 2015). Overall, it discovered that miR-197 contributed to the oncogenic activation of STAT3 via multiple targets through the upregulation of CKS1B.…”

“…Since miR-197 drove a highlight in cancer cell proliferation, had no significant influence in proliferation and cell cycle of colorectal cell lines and lung cancer cell lines (Fujita et al, 2015). Taken together, the functions of miR-197 in cell proliferation are not well known and the weight of miR-197 in cell proliferation deserves more researches.…”

“…Recently, Fujita et al showed that miR-197 regulates immune checkpoints in non-small cell lung cancers (61). Furthermore, miR-200/ZEB1 has been shown to control the expression of PD-L1 in tumors (62).…”

“…Furthermore, examination of clinical HCC samples showed that levels of miR-197 expression positively correlated with metastasis in HCC patients. Consistently, miR-197 has been shown to promote pulmonary metastasis for non-small cell lung cancer [37,46]. miR-197 also participates in the development of a number of types of cancer [26][27][28][29][30][31][32][33][34], including HCC [29,30], indicating the role of miR-197 in cancer.miR-197 also participates in liver fibrosis [35,36], in which Wnt/β-catenin signaling also plays a central role [47].…”
