The complete sequence of the mitochondrial genome of Saccharomyces cerevisiae

Abstract: The currently available yeast mitochondrial DNA (mtDNA) sequence is incomplete, contains many errors and is derived from several polymorphic strains. Here, we report that the mtDNA sequence of the strain used for nuclear genome sequencing assembles into a circular map of 85 779 bp which includes 10 kb of new sequence. We give a list of seven small hypothetical open reading frames (ORFs). Hot spots of point mutations are found in exons near the insertion sites of optional mobile group I intron-related sequences… Show more

“…In two clones we found the 21S rRNA at one extremity of the insert and the 15S rRNA at the other extremity. This shows that the 15S rRNA and 21S rRNA genes map close to each other (about 2 kb) on the mitochondrial map of S. kluyveri, in contrast to S. cerevisiae, in which at least 30.5 kb separate 15S from 21S rDNA [20]. The COX1 gene was identi¢ed in three RSTs.…”

“…When we started this work, only 46 S. kluyveri DNA sequences were available in GenBank, most of them corresponding to ribosomal RNA sequences (16) and a variety of nuclear genes (20). Only four of these sequences correspond to mitochondrial genes (COX3, COX2 and ATP8) and three to the pSKL plasmid.…”

“…The mitochondrial DNA of K. thermotolerans was previously characterized by restriction mapping [7]. With only 23 kb it is much smaller than the 86 kb S. cerevisiae mitochondrial genome [15]. We have found only 28 RSTs corresponding to mitochondrial DNA, 22 of them form seven small contigs.…”

“…First, a series of seven S. cerevisiae data ¢les comprising: (1) the 6213 predicted protein sequences as described in [1]; (2) the intergenic and subtelomeric DNA sequences [4]; (3) the 2 Wm plasmid DNA sequence [8]; (4) the rDNA sequences extracted from the chromosome XII sequence [9]; (5) the 52 distinct tRNA gene sequences as de¢ned in [1]; (6) the Ty elements DNA sequences [10]; and (7) the mitochondrial DNA sequence [11] and its translation products.…”
