The cytotoxicity evaluation of Kevlar and silicon carbide by MTT assay

Abstract: The MTT test has been widely used as a rapid and sensitive method for screening anticancer drugs. In this paper, we used this method to assess the cytocompatibility of three materials: Kevlar 29, silicon carbide and polyvinyl chloride (PVC) in both a quantitative and a qualitative manner. The materials were prepared by cleaning in 70% ethanol, autoclaved or gammasterilized. Extracts were prepared at four time periods (1, 2, 3 and 4 weeks) and two temperatures (37°C and 80°C). The extracts were used in the MTT … Show more

“…The results of the MTT-assay shown in the histogram in Figure 9, gives the optical density (OD) of the active cells in culture after 56 h. The assay detects metabolism in the living cells and the intensity obtained is dependent on the degree of activity of the cells. 32 The histogram (Figure 9) shows the result of the optical density obtained from viable cells in the wells coated with DLC and a-C:H:Si, indicating intracellular activity and/or proliferating potential of the endothelial cell in these samples. The uncoated TCPS was used as a control.…”

“…The MTT assay is a colorimetric assay which upon cleavage by viable cell's mitochondrial enzyme succinate-dehydrogenase yields a blue colored product formazan. The scanning multiwall spectrophotometer is used to read the amount of formazan crystals produced, which is proportional to the number of viable cells present (Mosmann 1983;Denizot and Lang 1986;Wan et al 1994). The breast cancer cell line MCF7 cells were obtained from the National Center for Cell Sciences, Pune, India.…”
