The Delipidation of Brain Proteolipid Protein by Ultrafiltration

Abstract: It has been very difficult to prepare the apoprotein moiety of brain white matter proteolipid so that it is completely devoid of complex lipids, without suffering aggregation and protein denaturation. The reason is that complex lipids are tightly bound to the proteolipid apoprotein. Using a new ultrafiltration method, we obtained, in a gradual way and in a relatively short time, more than 99% delipidation in water-saturated n-butanol, with and without 0.1 M acetic acid, and recovered up to 86% of the protein w… Show more

“…For delipidation, first a 10% SDS solution (w/v) was prepared and cooled in ice,29 and then 0.75 mL of this solution was added to the pellets to solubilize them. Then, 0.75 mL 12% TCA solution was added to the SDS‐solubilized pellets to a final TCA concentration of 6% and incubated at −20°C for 30 min.…”

“…Ten volumes of precooled benzene were added to a sample containing 5 mg proteolipid protein/ml organic mixture 1201. The lyophilised powder was solubilised at a constant concentration of 1 mg protein/ml organic mixture and purified by a delipidation procedure [21], in which an Amicon ultrafiltration cell, equipped with a 20000 MWCO membrane from Nuclepore Corp. (Pleasanton, USA) was used. Nitrogen at 0.1 MPa (1.5 lb/in2) and magnetic stirring were constantly applied to the cell.…”

“…We have obtained here an equivalent transition at 52 °C using a similar PLP purification procedure with organic solvents (Mateo et al 1986;Monreal 1975;Aguilar et al 1983). In our PLP preparations we found 14% total protein content, about 20% of which corresponds to DM-20 with no detectable MBP or Wolfgram fraction; that is to say, PLP represents about 80% of the total protein composition.…”
