The detection and estimation of Verticillium dahliae in naturally infested soil

Abstract: The effects of several factors on the estimation of Verticillium dahliae in soil by the wet-sieving method were studied. The following factors were important for maximising recovery: the removal of soil particles of < 20 nm size from suspensions before plating; the medium used for plating; the amount of sieved soil inoculated to plates; the length of time of incubating plates; and the method of incubating soil on plates. There was no short-term effect of air-drying soil before analysis. Using sodium hexametaph… Show more

“…For instance, it is advisable to avoid fields in close proximity with V. dahliae-susceptible crops, or to avoid soils where susceptible hosts are known to have been previously cropped. But most importantly, it is highly recommended to assess the presence of pathogen propagules in soil, either by traditional microbiological isolation procedures (Harris et al 1993;López-Escudero and Blanco-López 2001), or by molecular detection assays (Pérez-Artés et al 2005).…”

“…Traditional microbiological methods used to quantify V. dahliae MS in soil has been thoroughly reviewed by Harris et al (1993). These techniques involve the plating of a defined amount of soil, previously processed by diverse methods (i.e.…”

“…The plants were placed in a shifted double row (0.25 × 0.5 m). The soil was highly infested with V. dahliae (15.8 microsclerotia/g soil) which was determined by the wet-sieving method (Harris et al 1993).…”

“…The content of the bag filled with V. dahliae inoculum microsclerotia were ground, and 0.003 g of the material was brought onto a specific Verticillium medium (Modified soil extract agar (MSEA) (Harris et al, 1993) in threefold. Germinating microsclerotia were scored.…”
