The detection of peste des petits ruminants (PPR) virus antigen by agar gel precipitation test and counter-immunoelectrophoresis

Abstract: SUMMARYThe detectability of peste des petits ruminants (PPR) viral antigen in both ante-mortem secretions and necropsy samples from experimentally infected goats was investigated by both the agar gel precipitation test (AGPT) and counterimmunoelectrophoresis (CIE). Viral antigen was detected from 42'6 % of the samples tested by the AGPT and 80-3 % by CIE. The detection of viral antigen in a high proportion of the ocular and nasal secretions as well as the faeces and buccal scrapings, particularly from those co… Show more

“…Serological tests such as agar gel immuno-diffusion test (AGID)/AGPT [101], counterimmuno-electrophoresis (CIE) [101] and indirect ELISA, although popular in earlier days for tentative diagnosis, do not differentiate PPR and RP infections [98,104]. Advent of cell culture and molecular biological techniques has allowed development of specific, rapid and sensitive diagnostics.…”

“…A 1% (w/v) agar was prepared by boiling 1 g of agarose in 100 ml of barbitone acetate buffer. The CIEP protocol was essentially as described previously by Elhag Ali and Lees (1979), Majiyagbe et al (1984) and Obi and Patrick (1984).…”
