The development of an FIA-CD strategy for screening sulfated polysaccharides using antimalarial drugs and related species as probes

Abstract: Heterogeneous sulfated polysaccharides have attracted significant attention in light of their various biological activities. However, recent events involving heparin have dramatically illustrated that several analytical challenges exist in accounting for such species. In this case, tainted heparin was associated with acute reactions that lead to numerous deaths. Researchers were forced to use time-consuming, sophisticated techniques (e.g., enzymatic digestion, NMR, CE, HPLC, MS, etc.) to identify the cause of … Show more

“…Other ingenious methods for detection and quantification of OSCS in heparin include a heparinase inhibition assay (Aich et al, 2011), a convenient disposable strip-type electrochemical polyanion sensor (Kang et al, 2011), pyrolysis MS (Nemes et al, 2013), and spectropolarimetry (Chmielewski et al, 2011;Stanley et al, 2011).…”

“…Also, the use of the circular dichroism spectroscopy detection in an FIA system in UV/ Vis range, reported for screening of sulfated polysaccharides, may be mentioned as a rather unique application. 284 The development of a fast and reliable analytical method for determination of e.g. heparin is connected with the need to identify adulterants, which can be a source of severe allergic reactions, and heparin is now one of the most widely employed natural therapeutics available.…”
