The percentage of an oral dose of warfarin absorbed has been measured by two methods.Using the urine excretion of radioactivity following oral administration of HC warfarin in 4 sub;ects, between 61 % and 92% of an oral dose was recovered. This variation is due to differences in the duration of urine collection. Using a comparison of the area under the plasma concentration-time curve in 8 sub;ects, between 77.6% and 100% of an oral dose was apparently absorbed. The rate of warfarin absorption was rapid as ;udged by a consideration of the oral plasma concentration time curve in 8 sub;ects; peak drug concentrations in plasma were reached by 25 to 60 minutes after oral dosing. Rapid absorption was confirmed by measurements using two different pharmacokinetic model~. Absorption was complete by 120 minutes in 4 sub;ects.
