The Diverse Applications of Pancreatic Ductal Adenocarcinoma Organoids

Abstract: Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal solid malignancies. While immortalized cancer cell lines and genetically engineered murine models have increased our understanding of PDAC tumorigenesis, they do not recapitulate inter- and intra-patient heterogeneity. PDAC patient derived organoid (PDO) biobanks have overcome this hurdle, and provide an opportunity for the high throughput screening of potential new therapies. This review provides a summary of the PDAC PDO biobanks established t… Show more

“…Pancreatic ductal adenocarcinoma (PDAC) is among the most lethal malignancies, with a 5-year survival rate of less than 12% [ 1 ]. Many patients succumb to the disease within the first 6 months, reflecting late diagnosis, metastasis, and therapy resistance [ 2 , 3 ]. A distinctive feature of PDAC is the progression of early pancreatic intraepithelial neoplasia to an advanced unresectable disease [ 2 , 3 ].…”

A distinctive feature of PDAC is the progression of early pancreatic intraepithelial neoplasia to an advanced unresectable disease [ 2 , 3 ]. During this process, the histopathological phenotype of the tumor changes from low-grade, characterized by well-formed glandular epithelial structures, to high-grade, whereby epithelial cells are diffuse throughout the desmoplastic stroma [ 2 , 3 ].…”

“…Pancreatic cancer represents one of the most lethal solid tumor types; pancreatic ductal adenocarcinoma accounts for 90% of these tumors, and less than 5% are pancreatic neuroendocrine neoplasms [96,97]. Pancreatic cancer organoids have been generated to investigate tumor biology and promote clinical applications [96,[98][99][100][101][102][103]. PDOs derived from primary pancreatic ductal adenocarcinoma (PDAC) and matched liver metastases were generated [104].…”

“…A distinctive feature of PDAC is the progression of an early pancreatic intraepithelial neoplasia to advanced unresectable disease (Feldmann et al, 2007). During this process, the histopathological phenotype of a tumor changes from low grade, characterized by well-formed glandular epithelial structures, to high grade, whereby epithelial cells are diffuse throughout the desmoplastic stroma (Low et al, 2021; Schlitter et al, 2017). This process of dissemination has been linked to the activation of epithelial-mesenchymal transition (EMT), a cell-biological program that is essential to the early stages of embryogenesis, the formation of organs and exploited by cancer cells (Thiery, 2002; Yang et al, 2020).…”
