The Drug Repurposing Encyclopedia (DRE): a web server for systematic drug repurposing across 20 organisms

Abstract: The identification of new therapeutic uses for compounds via computational or experimental approaches, which is widely known as drug repurposing, has the potential to develop novel therapies with pre-existing medicines, thereby reducing the time and costs associated with drug development. Today, several data-driven methodologies have been developed leading to databases that facilitate drug repurposing initiatives. However, no approach has systematically compared drug transcriptional profiles to those from a wi… Show more

“…gov/geo/geo2r/ accessed on 23 December 2023] prior to searching for potential drugs. The Drug Repurposing Encyclopedia (DRE) webserver was used to search for available drugs that can be repurposed based on their transcription profiles from the Molecular Signatures Database (MSigDB) and Connectivity Map (CMap) [24,25]. The groups of upregulated and downregulated genes were submitted for drug repurposing tests based on molecular signature screening.…”

“…The two groups were then fed to the Drug Repurposing Encyclopedia (DRE) web server, where they were subjected to drug repurposing. The DRE [https://www.drugrep.org/drugrepurposinganalysis (accessed on 10 January 2024)] is an online drug discovery platform that is referenced from four large drug databases intended for drug repurposing analyses, screening over a total of 4690 consensus drugs from 20 studied organisms [25]. The DEGs were treated as gene signatures and inputted into the DRE.…”

“…Screening for repurposeable drugs was performed using the recently launched Drug Repurposing Encyclopedia (DRE) (https://www.drugrep.org (accessed 18 May 2023)), an interactive web server that makes use of over 198 million significant drug-signature associations from the Molecular Signatures Database (MSigDB) and over 30,000 drugassociated transcription profiles from Connectivity Map (CMap) [39]. The top ten (10) hub genes in each module were used as gene signatures and grouped into upregulated and downregulated based on differential expression analysis (DEA) using GEO2R (https: //www.ncbi.nlm.nih.gov/geo/geo2r (accessed 18 May 2023)) before being submitted to the DRE webserver for drug repurposing analysis.…”
