“…However, there were a greater number of blast cells (5.1±2.9%) in the cultures which had been exposed to HgCl2 for the full 6 day culture period. Because of the evidence that granulocytes and possibly macro phages play a role in specific antigen induced lymphocyte transforma tion [10,11] we examined the response of column purified lymphocytes to HgCl.>. An aliquot of leucocyte-rich plasma was allowed to stand on a glass wool column at 37° C for 1 h and was then eluted with autologous leucocyte free plasma.…”

“…This hypothesis might explain why other workers failed to obtain lymphocyte transformation despite marked cellular disparity when homol ogous or heterologous cells of solid tissue origin were used as stimulants [8,15] inasmuch 'mediator' macrophages would have been lacking in the allogeneic cell suspensions. This situation would be different in mixed leucocyte cultures where macrophages are thought to arise from cells having the appearance of small lymphocytes [5,7,11]. Furthermore the finding that macrophages after disruption by ultrasonication loose this enhancement effect supports the concept of intercellular immune transfer depending on viable macrophages.…”

“…They are probably numerous [6,10,11], however, since even in labora tories performing this test under standardized conditions repetitively the MLC does not provide results with exact reproducibility.…”

“…G o u g h , E lves and I sraels (8) have shown that these macrophages are derived from small lympho cytes and that their occurrence is related in some way to the pre sence of polymorphonuclear leucocytes. In a systematic study of the effect of polymorphonuclear leucocytes on transformation in mixed lymphocyte cultures J ones (10) has confirmed that the num ber of phagocytic macrophages which appear in both control and stimulated leucocyte cultures depends on the number of polymor phonuclear leucocytes originally present. O ppen h eim and W o lff ( 16) have recently shown by autoradiography that in lymphocyte trans formation induced in vitro by carbon 14 labelled Salmonella enteritidis endotoxin the antigen was taken up by macrophages and not by the lymphocytes which were stimulated to transform.…”
