The effect of the DLTIDDSYWYRI motif of the human laminin α2 chain on implant osseointegration

Abstract: Considerable effort has been directed towards replacing lost teeth using tissue-engineering methods such as titanium implants. A number of studies have tried to modify bioinert titanium surfaces by coating them with functionally bioactive molecules for faster and stronger osseointegration than pure titanium surfaces. Recently, peptides have been recognized as valuable scientific tools in the field of tissue-engineering. The DLTIDDSYWYRI motif of the human laminin-2 α2 chain has been previously reported to prom… Show more

“…DLTIDDSYWYRI is an active motif from the large globular 1 domain of human laminin-2 a2 chain that promotes osteoblast differentiation [157,158]. DLTIDDSYWYRI acts through syndecan-1 on the cell membrane resulting in phosphorylation of downstream protein kinase C (PKC) delta leading to cell adhesion and enhanced osteointegration of implants coated with the peptide in vivo [158,159]. GFOGER is a peptide which resembles sequences on the collagen I α1(I) chain.…”

“…One type of laminin‐derived peptide is the DLTIDDSYWYRI motif (also known as Ln2‐P3), which originates from the human laminin α2 chain. Bone healing is markedly accelerated when commercially pure titanium (Ti) dental implants are treated with Ln2‐P3 (Kang et al, ). The mechanism by which this acceleration occurs involves syndecan‐1, which is a transmembrane proteoglycan receptor expressed on osteoblasts.…”

“…Recombinant human bone morphogenetic protein 2 (rhBMP-2) coated on an oxidized Ti surface enhanced the bone affinity and healing capacity of the surface, compared with the oxidized Ti surface only [67]. Furthermore, several short peptides, derived from cell adhesion proteins, have been effective in bone cell attachment and are non-antigenic to implant surfaces [70,71]. Both in vitro and in vivo experiments show promising results in terms of biologic responses surrounding protein or peptide-coated implants [71,72].…”

Both in vitro and in vivo experiments show promising results in terms of biologic responses surrounding protein or peptide-coated implants [71,72]. A laminin-derived peptide showed the increased expression of the osteogenic markers at the gene level, the enhanced alkaline phosphatase activity at the protein level, the improved attachment and spreading of osteoblast-like cells, and the accelerated bone response in the rabbit tibia model [71].…”
