The efficacy and safety of omega-3 fatty acids on depressive symptoms in perinatal women: a meta-analysis of randomized placebo-controlled trials

Abstract: Omega-3 fatty acids (FA), as a nutrient, has been proven effective in major depressive disorder (MDD), however, the results of monotherapy in perinatal depression (PND) remain unclear. To examine the efficacy and safety of omega-3 fatty acids (FA) monotherapy for perinatal depression (PND) compared with placebo. PubMed, Embase, PsycINFO, MEDLINE, Cochrane Library, and CINAHL were searched from inception up to November 2019. The reference lists of relevant review articles and included studies were also reviewed… Show more

“…FAs are then liberated inside the cytosol and via cyclooxygenase-2 (COX-2), 5-lipoxygenase and thromboxane synthetase activity gets degraded into PGs, LTs, and TX respectively. Thus, omega-3 FA is responsible for decreasing the synthesis of eicosanoids derived from arachidonic acid (AA) like PGI2, PGF2α, PGE2, and LTB4 which are responsible for pro-inflammatory responses and increase the synthesis of LT5 and PGE3 from EPA which have weak effects on inflammation [ 57 58 ].…”

“…Some mild gastrointestinal symptoms such as nausea, vomiting, loose stools, and increased stool frequency may be associated with omega-3 FAs. Few patients might report neurologic symptoms such as dizziness and insomnia; however, most symptoms are very mild and self-limiting [ 50 57 ]. Elderly people are at risk of bleeding when they combine long chain PUFA such as fish oil with other anticoagulants such as warfarin and aspirin.…”

“…There is little doubt that a low Omega-3 Index during pregnancy is concentration-dependently associated with risk for perinatal depression [ 42 , 43 , 44 ]. Increasing dietary intake of EPA and DHA, preferably with a product containing more EPA than DHA, is safe in pregnant women and improves perinatal depression, although not all meta-analyses agree on the efficacy, probably illustrating the methodologic issues discussed above [ 45 , 46 , 47 , 48 ].…”

“…Our findings show that HT or injection of tri-DHA emulsion reduce infarct volume and the degree of neuroprotection is similar for both treatments. Omega-3 fatty acids are safe and well tolerated in humans without major adverse effects ( 64 66 ). Intravenous injections are a common feasible procedure, which can be easily performed in primary care settings.…”
