The EWSR1/NR4A3 fusion protein of extraskeletal myxoid chondrosarcoma activates the PPARG nuclear receptor gene

Abstract: The NR4A3 nuclear receptor is implicated in the development of extraskeletal myxoid chondrosarcoma (EMC), primitive sarcoma unrelated to conventional chondrosarcomas, through a specific fusion with EWSR1 resulting in an aberrant fusion protein that is thought to disrupt the transcriptional regulation of specific target genes. We performed an expression microarray analysis of EMC tumors expressing the EWSR1/NR4A3 fusion protein, comparing their expression profiles to those of other sarcoma types. We thereby ide… Show more

“…The 739 probe sets on the U133A chip corresponded to 432 genes with kinase domains, representing 83% of the 518 known protein kinase genes. (Note: this microarray expression analysis was utilized in a previous publication as a comparator data set for a study of myxoid chondrosarcoma (26)).…”

“…Also, a t(9;17)(q22;q11) and a t(9;15) (q22;q21) are found, fusing the NR4A3 gene with RBP56 and TAF15, respectively. Recent data indicate that the EWSR1/NR4A3 fusion protein may activate the PPARG nuclear receptor gene that may not only play a key role in the molecular oncogenesis of EMCS but also represents a potential therapeutic target [100].…”

“…To determine whether neural genes are enriched in primary DSRCT, we analyzed the expression profile of 28 primary DSRCT performed previously in comparison with four other tumor types (31). GO analysis of genes that are upregulated (>2-fold) in DSRCT compared with either Ewing sarcoma (ES), alveolar rhabdomyosarcoma (ARMS), or alveolar soft part sarcoma (ASPS) revealed an enrichment of neural pathways (Supplementary Table S2), but not when compared with synovial sarcoma (SS).…”
