The Expression of Melanoma-Associated Antigen D2 Both in Surgically Resected and Serum Samples Serves as Clinically Relevant Biomarker of Gastric Cancer Progression

Abstract: MAGE-D2 was associated with metastatic potential of GC and may represent a promising biomarker, both in gastric tissues and serum samples, for malignant behavior of GC.

“…Although palliative chemotherapy and surgery are beneficial, the outcome of GC remains dismal (4)(5)(6). The high mortality of GC is explained by our poor understanding of its mechanism of progression and the lack of suitable diagnostic markers that hinder diagnosis before the disease reaches an advanced stage (7,8). GC represents a biologically and genetically heterogeneous group of tumours that are induced by multiple factors that deregulate cell signalling pathways, which leads to the acquisition of malignant phenotypes such as increased cell proliferation, inhibition of apoptosis and enhanced invasiveness (9)(10)(11).…”

“…Furthermore, the identification of molecular biomarkers that predict treatment efficacy will likely improve our ability to manage these patients. 22 It is important to note that the outcome of the COMET trial is highly significant because of its data regarding safety and influence on PFS after liver resection. For example, the overall frequency of grade 3/4 AEs, including hematologic and nonhematologic events, was 45 %, which is generally consistent that of the new EPOC trial (preoperative cetuximab plus chemotherapy, 40 %) and an earlier clinical trial conducted in the United Kingdom.…”

“…By contrast, the profile of prognostic factors was dramatically altered in patients who received S-1, with macroscopic tumor size ‡50 mm identified as the only significant prognostic factor [68]. A large body of evidence indicates that the depth of tumor invasion serves as an important prognostic factor for gastric cancer as a major determinant of tumor node metastasis staging, partially because serosal infiltration correlates with peritoneal recurrence [2,69,70]. Ultimately, these results emphasize that new methods for patient stratification will need to be established in patients who are receiving more intense treatments, such as curative gastrectomy followed by adjuvant chemotherapy, with the risk for recurrence.…”
