The Fate of Biogenic Monoamines in Perfused Rabbit Lung

Abstract: I Inactivation of f3-phenylethylamine and several of its derivatives was studied in a preparation of rabbit lung perfused with Krebs physiological medium at 370C. Inactivation or removal of these compounds was calculated as the difference between the concentration of each amine in the perfusion medium and the effluent, collected separately from each lung. The extent of amine metabolic degradation was also measured, by column chromatography, in lung effluent. 2 With this technique the magnitude of amine removal… Show more

“…On the other hand, PHE metabolism in the perfused lung has an apparent Km of 54 gIM which is very close to that measured in vitro. In rabbit isolated lung, PHE removal was anomalous in that alone out of six substrates the removal of this amine showed no saturation up to 830 gM, whereas 5-HT and noradrenaline in particular reached saturation at about 3 to 6 jiM (Gillis & Roth, 1977). The lack of effect of desmethylimipramine on PHE metabolism was unexpected for two reasons.…”

We had therefore expected desmethylimipramine to decrease PHE metabolism, if not by inhibiting MAO, then at least by inhibiting uptake.…”

“…MID methods applied to the lungs have the potential for discriminating among lungs having different physical and chemical properties such as occur in lung injury and disease (7,8,22,23,28,29,35). This is the basis for MID studies of lung water volume (8,12,21,28), capillary permeability (7,8,14,15,22,23,36,38), endothelial enzyme activity (8,9,15,18,29,35), and other aspects of lung tissue function (1-5, 7, 8, 16, 19, 20).…”

“…This was followed by distilled water (3 mL) and the eluates containing 5-HIAA were collected in pre-weighed glass vials. The adsorbed 5-HT was subsequently eluted with 0Á5 M hydrochloric acid (3 mL) into other pre-weighed glass vials (Gillis & Roth 1977). The vials were re-weighed and the volume of each eluate was determined.…”
