Pepper vein yellows virus (PeVYV) is a newly recognized Polerovirus extracted from Chinese pepper. The symptoms of PeVYV-infested pepper plants comprise intervein yellow staining, leaf curl formation and other malformations, and leaf internodal shrinkage, but the roles of the viral proteins remain undetermined. The P0 protein of the genus Polerovirus has established post-transcriptional gene silencing (PTGS) activity. This investigation focused on the PeVYV-encoded P0 protein and assessed its potential virulence capacity, PTGS activity, and tendencies to localize in the nucleus. This study revealed that P0 influenced the pathogenic properties of a specific heterologous potato virus X. In addition, P0 proteins impaired local gene silencing, although they did not regulate generalized gene silencing within Nicotiana benthamiana 16c plants. Furthermore, P0 proteins localized mainly in the nucleus, particularly in the nucleolus. P0 deletion mutagenesis demonstrated that the F-box motif (56–72 amino acids, AAs) of P0 was essential for symptom determination, inhibition of PTGS, and subcellular localization. Mutation analysis of the F-box motif of P0 protein indicated that AA 57 of the P0 protein was a pivotal site in symptom development and that AA 56 of the P0 protein was indispensable for inhibiting PTGS and subcellular localization. The outcomes obtained here suggest that further studies should be conducted on the molecular mechanisms of amino acids of the F-box domain of P0 protein in the interaction of PeVYV with plants.

Since their first observing in Japan, poleroviruses have threatened pepper cultivation all around the World. Symptoms of vein yellowing originating from poleroviruses have become widespread in both open field and protected pepper cultivation areas recently. In this work samples collected from highly infected pepper fields in South East Turkey have been determined as Pepper Vein Yellows Virus (PVYV) and Beet Western Yellows Virus (BWYV) (genus Polerovirus, family Luteoviridae). Recombinant PVYV isolates have been observed from intra and interspecific combination in relation to part of the RNA-dependent RNA polymerase coding region subjected to recombination detection methods. One of the PVYV isolates has induced recombination between BWYV and PVYV and located middle of the PVYV and BWYV isolates. Another isolate recombined from intraspecific combination has been transferred onto pepper genotypes carrying pvr alleles and landraces under both artificial and natural conditions to determine their response. None of the pvr alleles in pepper genotypes used in this study could contribute to resistance to PVYV isolates.

Pepper vein yellows virus (PeVYV, Luteovirdae; Polerovirus) infects Capsicum plants, whose bell pepper fruits (Capsicum annuum) are important crops in Japan. Reductions in the yield and quality of bell pepper fruits caused by PeVYV are agricultural problems in Okinawa Prefecture, Japan. The prevalence of PeVYV deposits in the annual disposition of harvested bell pepper plants suggests the possible existence of a reservoir for the virus. Here, we examined bird pepper plants (Capsicum frutescens) that were grown wild on five islands in Okinawa Prefecture, as well as cultivated peppers (C. annuum, Capsicum baccatum and C. frutescens) for the presence of PeVYV by reverse transcription polymerase reactions using specific primers. Overall, PeVYV was widely detected in bird peppers on the three islands (Okinawa, Miyako and Yonaguni) where the bell pepper disease occurred, but not detected in the plants from two islands (Ishigaki and Minami-Daito) with no disease. Laboratory experiments demonstrated that an Okinawa-derived strain of cotton aphid Aphis gossypii transmitted PeVYV from viruliferous wild bird peppers to the bell peppers, which may suggest an annual supply of PeVYV from wild bird peppers to the bell peppers via aphids in Okinawa Prefecture. Molecular phylogenetic analyses based on the nucleotide sequences of seven distinct genomic regions suggested that PeVYV detected from the bell peppers and bird peppers are indistinguishable, supporting the view that PeVYV moves frequently between these host plants.
