The genome sequence of a caddisfly, Limnephilus lunatus (Curtis, 1834)

Abstract: We present a genome assembly from an individual Limnephilus lunatus (a caddisfly; Arthropoda; Insecta; Trichoptera; Limnephilidae). The genome sequence is 1,270 megabases in span. Most of the assembly is scaffolded into 13 chromosomal pseudomolecules, including the assembled Z chromosome. The mitochondrial genome has also been assembled and is 15.4 kilobases long.

“…In this study, we report eight new full-length h-fibroin sequences for caddisflies across a diverse set of silk usage. Two of these were generated from new genomic resources, whereas five were mined from previously published genomes, 41 , 42 , 43 , 44 highlighting the relevance of genome sequencing projects for the wider scientific community. The new full-length h-fibroins represent a substantial increase in the number of genomic resources available for the study of caddisfly silk and allowed us to compare the major silk gene for eleven species across the primary clades of Trichoptera, in which species exhibit different silk usage.…”

“…We manually curated the transcript sequences of the candidate silk genes in L. lunatus against the available genomic sequences (GenBank:GCA_917563855.2) [ 27 ] using BLAST searches [ 28 ]. For easier distinguishing of exon–intron boundaries and visualization of genome clusters, we mapped the Illumina reads to the genome using RNA STAR software (Galaxy version 2.7.8a + galaxy0) in combination with IGV 2.9.4 [ 20 ].…”
