The host-range, genomics and proteomics of Escherichia coli O157:H7 bacteriophage rV5

Abstract: BackgroundBacteriophages (phages) have been used extensively as analytical tools to type bacterial cultures and recently for control of zoonotic foodborne pathogens in foods and in animal reservoirs.MethodsWe examined the host range, morphology, genome and proteome of the lytic E. coli O157 phage rV5, derived from phage V5, which is a member of an Escherichia coli O157:H7 phage typing set.ResultsPhage rV5 is a member of the Myoviridae family possessing an icosahedral head of 91 nm between opposite apices. “…Five to 6 thin tail fibers of 61 nm in length were occasionally seen. The morphology of AXO121A closely resembles that reported for phage rV5, in which the head measured 91 nm in diameter and the contractile tail was 121 nm by 17 nm with 70 nm tail fibers (Kropinski et al 2013). Phages AYO26A and vB_EocM_AXO121B had a large elongated head with a diameter of 115-117 nm.…”

“…The host range of phage Bp7 for 35 clinical E. coli strains and 4 laboratory E. coli strains was determined by spotting 10 5 PFU of Bp7 onto freshly seeded lawns of bacteria on agar plates (13). Each test was repeated three times.…”

“…They share (1) genome sizes ranging from 136 kb (phage vB_ EcoM-FV3) to 148 kb (phage phi92), (2) similar number of protein coding sequences (CDSs) ranging from 233 (rV5) to 269 (vB_ CsaM_GAP31), (3) mol.% G + C ranging from 37.4 % (phi92) to 46.3 % (vB_CsaM_GAP31), and (4) number of tRNAs ranging from fi ve (vB_EcoM-FV3) to 26 (vB_CsaM_GAP31) [ 36 -41 ]. Recently, doubt has been cast on whether this grouping represents a genus or subfamily ("V5virinae") containing three genera-the "V5likevirus" (rV5, FV3), the "Pvplikevirus" (PVP-SE1, GAP31, and SSE-121), and the "Phi92likevirus" (phi92 and phAPEC8) [ 37 ].…”
