The Caenorhabditis elegans gene unc-76 and its human homologs define a new gene family involved in axonal outgrowth and fasciculation

Abstract: The gene unc-76 (unc, uncoordinated) is necessary for normal axonal bundling and elongation within axon bundles in the nematode Caenorhabditis elegans. The UNC-76 protein and two human homologs identified as expressed sequence tags are not similar to previously characterized proteins and thus represent a new protein family. At least one of these human homologs can function in C. elegans, suggesting that it, like UNC-76, acts in axonal outgrowth. We propose that the UNC-76 protein, which is found in cell bodies… Show more

“…Loss of UNC-76 function in nematodes results in defective axonal transport and locomotion that can be rescued by FEZ1. 204 In rat brain, FEZ1 is expressed primarily in neurons in a pattern closely paralleling that of DISC1, 205 and in rat hippocampal neurons reduced FEZ1 expression induced by RNA interference results in several neuronal defects affecting neuronal polarity and axon growth, as well as intracellular transport. 206 The latter observation is consistent with demonstrations of an involvement of FEZ1 in kinesin-based motility.…”

“…Fire) or an unc-76 promoter vector (Bloom and Horvitz, 1997) to generate myo-3::sup-9 and unc-76::sup-9, respectively. The n1550 mutation was introduced into myo-3::sup-9 using mutagenic PCR primers.…”

“…One of the positive clones encoded a partial sequence DISC1 participates in neurite outgrowth K Miyoshi et al of FEZ1 (amino acids 129-392), which is a mammalian homolog of the Caenorhabditis elegans UNC-76 protein involved in axonal outgrowth and fasciculation. 21 UNC-76 and FEZ1 are not similar to any previously characterized proteins and represent a new protein family. 21 Human FEZ1 protein is able to complement the function of UNC-76 in the nematode.…”
