“…To assess this hypothesis, we evaluated the changes in TEER values on CaCo-2 cells after 1, 2, 3, and 4 weeks of culture time. As reported in Figure S2 (Lea, 2015;Olejnik et al, 2003;Sambuy et al, 2005;Ye et al, 2015). A similar morphological profile was exhibited when they were seeded on the 3D-CGE (Figure 4b).…”

“…Human colonic epithelial cell line, Caco-2 cells (passage #25-35), were obtained from American Type Cell Culture (ATCC, Manassas, VA) and were directly seeded on tissue culture polycarbonate membrane filters in 24-well Transwell ® plates (pore size 3.0 μm, Corning, New York, NY) (Kowapradit et al, 2010;Lea, 2015;Schimpel et al, 2014;Svensson, Finlay, Bass, von Bonsdorff, & Greenberg, 1991;Ye, Dawson, & Lynch, 2015) at a seeding density of 1 × 10 4 cells/cm 2 (Kowapradit et al, 2010;Olejnik, Lewandowska, Grajek, & Czaczyk, 2003), in order to produce 2D transwell culture. For organotypic culture, 3D-CGE and 3D-CSSE (6 mm diameter) were rinsed with three washes of PBS solution, then were accommodate in the 24well Transwell ® inserts (diameter, 6.5 mm; Corning) and allowed to dry for 5 min under laminar flow.…”

“…We first examined the barrier properties of two epithelial cell lines Caco-2 (colon) and A549 (lung) by growing these cells on transwell inserts for 4 days and measuring the trans-epithelial electrical resistance (TEER) every day. Caco-2 cells have been reported to have higher expression of tight junction proteins and undergo differentiation whereas A549 cells have lower TEER values and do not undergo differentiation (27)(28)(29)(30). As expected, the basal levels of TEER was about 200-fold lower in A549 cells as compared to Caco-2 on day 2, however, both cell lines showed an increase in the TEER values with time in culture and started to plateau by day 4 (Supplementary Figures 1A,B).…”

“…Static models differ in several important aspects: type and concentration of enzyme used, number of steps and time of particular phase of digestion, rate of stirring, and other parameters (Hur, Lim, Decker, & McClements, 2011). Dynamic models include division into subcompartments, mimicking different structures and functionalities in the gastrointestinal tract, and account for physical forces and changes in kinetics during digestion (Thuenemann, 2015 How amino acid sequence of peptide influences biological activity . .…”
