The influence of diet consistence, drinking water and bedding on periodontal disease in Sprague‐Dawley rats

Abstract: The results of the present study emphasize the need for pre- experimental examination of rats. They also show that diet and bedding conditions have the potential of seriously influencing outcomes of studies of periodontal disease in rats.

“…16 Therefore, ovariectomy-induced osteopenia in the rat produces skeletal responses similar to those in post-menopausal women. 17 A morphometric analysis of defleshed jaws [11][12][13]18,19 was applied to assess the amount of periodontal bone in the rats. This analysis is reproducible and can discriminate among experimental groups.…”

“…The total alveolar bone loss was obtained by taking the sum of the recordings from the buccal tooth surface and subtracting the value of the right maxilla (unligated control) from the left, in millimeters. Although it is claimed that laboratory rats may harbor ongoing baseline periodontitis (23), in the present study we used the contralateral right maxilla as the unligated control in order to avoid the influence of a baseline periodontitis on our results.…”

“…To avoid spontaneous PD (Bjornsson et al, 2003), the animals were bred on Teklad 7089 Diamond Soft Bedding (Harlan Laboratories, Livermore, CA, USA). The rat pups were placed on the special bedding before they were 15 days old, with free access to food (Purina #5008, LabDiet, St. Louis, MO, USA) and water.…”

“…Before PD was induced, all rats were examined to exclude rats with pre‐existing PD (defined by probing depths >0.5 mm) (Bjornsson et al, 2003). Each week, PD induction was checked under general anesthesia and loose or lost ligatures were replaced as previously described (17; 18).…”
