The influence of LTS-4, a saponoside from Lysimachia thyrsiflora L., on human skin fibroblasts and human melanoma cells

Abstract: Abstract:We investigated the effect of a triterpene saponoside from Lysimachia thyrsiflora L. upon the viability, proliferation, morphology and cell motility of human melanoma HTB-140 cells and human skin fibroblasts (HSFs). The compound, denoted LTS-4, decreased the viability and rate of cell growth of both cell types in a time-and dose-dependent manner, and proved cytotoxic against cancer cells at significantly lower concentrations than for fibroblasts. LTS-4 also affected the morphology of the examined cell… Show more

“…Figure demonstrates the composite trajectories of HSFs migrating on the glass surface noncovered (control) and covered with the various polyelectrolytes. The trajectories are shown as circular diagrams with the initial point of each trajectory placed at the origin of the plot as previously described . The average motility parameters calculated from the cell tracks are presented in Table .…”

“…The trajectories are shown as circular diagrams with the initial point of each trajectory placed at the origin of the plot as previously described. 22,32 The average motility parameters calculated from the cell tracks are presented in Table III. The final cell displacement shows the migration efficiency of cells into a wound area.…”

“…The trajectories of individual cells were evaluated from the changes in cell centroid location, as described previously [ 18 ]. For each cell, the following variables were determined [ 19 ]: (i) average speed of cell movement, i.e. the total length of cell trajectory/time of recording; (ii) the total length of cell displacement (μm), i.e.…”

“…In cancer therapy, they were applied in combination with immunotoxins to increase the selectivity of the immunotoxin against cancer cells and to inure normal cells to its cytotoxic effect (Weng et al, 2012). On the other hand, direct pro-apoptotic, antiangiogenic, cytostatic and anti-invasive activity of saponosides has been demonstrated (Galanty et al, 2008;Kim et al, 2012;Podolak et al, 2013). They can enhance the effect of commonly used chemotherapeutic agents, suggesting possibilites for lowering their therapeutic doses and reducing the possible side-effects of chemotherapy The observations of therapeutic efficacy of the combinations of some natural products from Traditional Chinese Medicine with standard chemotherapeutic agents (Efferth et al, 2002;Hofman et al, 2012) warrant intensive research on the anti-cancer activity of saponosides and on the mechanisms of their interference with chemotherapeutics.…”
