The Inhibitory Mechanisms Study of 5,6,4′-Trihydroxy-7,3′-Dimethoxyflavone against the LPS-Induced Macrophage Inflammatory Responses through the Antioxidant Ability

Abstract: Abstract:The whole plant of Anisomeles ovata has been widely used in Taiwan for treating inflammation-related skin and liver diseases, however, the detailed pharmacology mechanisms have yet to be elucidated. In the present study, one of the major components, 5,6,4 1 -trihydroxy-7,3 1 -dimethoxyflavone (5-TDMF), was purified from a methanol extract of Anisomeles ovata. A pharmacological study of this compound suggests that 5-TDMF possesses potent free radical scavenging activity both in vitro and ex vivo. Furth… Show more

“…The same study suggested that (5-TDMF) could block LPS-induced NF-κB translocation and iNOS and COX-2 expressions by inhibiting the mitogen-activated protein (MAP) kinase and MAPK/ERK signaling pathways. Suggesting potential novel chemo-preventive anti-inflammatory agents [ 40 ]. Still, a lack of detailed studies is noticed regarding its molecular mechanism that controls its activities, such as binding to a particular protein structure [ 65 ].…”

“…In addition to these two compounds, quercetin-pentamethyl ether and trihydroxy-dimethoxyflavone were also present in KP-SeNPs. Quercetin-pentamethyl ether was found at its highest peak in KP-SeNPs, which showed sirtuin-activating and anti-glycation activities in a previous report [ 67 ], and trihydroxy-dimethoxyflavone showed potent anti-inflammatory activity without any observable cytotoxicity [ 68 ]. Thus, we speculate that these two compositions may contribute to the good biosafety of KP-SeNPs.…”

“…[8] Certain compounds, such as nevadensin, chlorogenic acid, 5, 6, 4'-trihydroxy-7, 3'-dimethoxyflavone, and syringic acid have been reported to exhibit antioxidant, anti-tubercular, antimicrobial, anti-inflammatory, anti-diabetic and anticancer activities. [25][26][27][28] Apiin, a natural flavonoid glycoside, exerts antiproliferative and apoptotic effects on human cancer cells. [29] Ladanetin, a dimethoxyflavone, is an effective anti-HCV agent, plant metabolite, antiviral agent, and radical scavenger.…”
