The intracellular carboxyl terminal domain of Vangl proteins contains plasma membrane targeting signals

Abstract: Vangl1 and Vangl2 are integral membrane proteins that play a critical role in establishing planar cell polarity (PCP) in epithelial cells and are required for convergent extension (CE) movements during embryogenesis. Their proper targeting to the plasma membrane (PM) is required for function. We created discrete deletions at the amino and carboxy termini of Vangl1 and monitored the effect of the mutations on PM targeting in Madin-Darby canine kidney cells. Our results show that the Vangl1 amino terminus lacks … Show more

“…In addition, immunofluorescence analysis showed that TMEM72 colocalized with the plasma membrane marker Na + ‐K + ATPase (Fig. 2D) [19], indicating the plasma membrane localization of TMEM72. Further microscopic analysis showed the intense localization of TMEM72 at cell–cell junctions (Fig.…”

“…Many of the domains critical for the scaffolding properties of Vangl are located within the large cytoplasmic C-terminal tail. Both Vangls have conserved basolateral-sorting motifs (YXXF) critical for the recruitment of Vangl into transport vesicles, and proper cell membrane localization of Vangl ( Guo et al, 2013 ; Iliescu and Gros, 2014 ). More recently, a p97/VCP-interacting motif (VIM motif) was identified in the C-terminal of Vangl2 that is required for direct interactions between Vangl2 and p97/VCP, a chaperone-related ATPase, which is critical for ubiquitination and degradation of Vangl through the endoplasmic reticulum-associated degradation (ERAD) pathway ( Feng et al, 2021 ).…”

“…The variant, interpreted as possibly damaging and deleterious by Polyphen and SIFT, respectively, is relatively frequent in the African population in which it is also present in the homozygous state, while it is extremely rare in other populations (0.0001238 among NFE to which the patient belongs: gnomAD). The variant is found in the carboxylic terminal of the protein, essential for its binding to the plasma membrane to make it available to the other PCP core proteins (Iliescu 2014 ). The functional study of an NTD variant that fell within the same domain (p.Arg374His) showed that the protein localized throughout the cytoplasm, rather than being enriched in the plasma membrane.…”
