The Irinotecan/5-Fluorouracil Combination Induces Apoptosis and Enhances Manganese Superoxide Dismutase Activity in HT-29 Human Colon Carcinoma Cells

Abstract: Background: We examined whether induction of apoptosis and Mn-superoxide dismutase (Mn-SOD) and Cu,Zn-superoxide dismutase (Cu,Zn-SOD) activities were involved in the greater cytotoxicity of the irinotecan (CPT-11)/5-fluorouracil (5-FU) combination for human colon cancer cells when compared to both drugs alone. Methods: HT-29 and SNU-C4 human colon carcinoma cell lines were treated with 5-FU and CPT-11, then apoptosis was evaluated by flow cytometry and SOD activities were determined by polyacrylamide gel elec… Show more

“…With the APH assay, classic drug combinations such as 5-FU and Irinotecan, which already showed increased response rate, time to progression, and survival in patients, 50 can now be more easily monitored in spheroids and compared with monolayer data. 38,[51][52][53] A particular prospective is the potential application of the spheroid-APH assay platform for the testing of single and combined therapeutic strategies with new, target, and/or pathway-specific treatment modalities. Further experiments to evaluate the feasibility of this approach are in progress.…”

“…The immunopositivity, however, was found to be rare in normal colorectal epithelium. The higher frequency of LC3 expression in colorectal adenocarcinomas compared with the corresponding normal tissue may indicate that the ex- each pathway may play an important role in cell death, which is associated with chemotherapy regimens (3,5,8,13,16). Further investigation is therefore needed to clarify the contribution of CC8, CC9, AIF, and LC3 in gastrointestinal cancer prognosis or response to treatment.…”

“…Recently, Grivicich et al [14] reported that CPT-11 and 5-FU combination therapy induces apoptosis. Oxaliplatin, the key drug of FOLFOX, is mainly excreted into urine.…”
