The Landscape of Pseudomonas aeruginosa Membrane-Associated Proteins

Abstract: Background: Pseudomonas aeruginosa cell envelope-associated proteins play a relevant role in infection mechanisms. They can contribute to the antibiotic resistance of the bacterial cells and be involved in the interaction with host cells. Thus, studies contributing to elucidating these key molecular elements are of great importance to find alternative therapeutics. Methods: Proteins and peptides were extracted by different methods and analyzed by Multidimensional Protein Identification Technology (MudPIT) appr… Show more

“…Proteomic data were obtained as previously described 9 . The identified proteins were ranked based on their abundance estimated by normalized peptide spectrum match (nPSM) 31 , whereby PSM was normalized for protein length by applying the formula nPSM=PSM/MW(KDa)*100.…”

“…To evaluate the correlation among replicate analyses, Spearman's rank correlation was computed by JMP15.2 SAS software. To provide a rank list of the most abundant proteins in the analyzed samples, the average Spectral count (SpC) of each protein was normalized on MW as previously reported [53]. Functional Annotation Tool of DAVID database [54] was used to characterize the most enriched molecular function (MF), biological process (BP), and cellular component categories; specifically, background = Equus caballus, count > 5 and p < 0.01 were set.…”

“…The dicationic compounds showed different activities: the porphyrins, P12 and P13, bearing substituents with longer chains (4 carbon and 8 carbon) in 5 and 15 positions were toxic, while P11 bearing a benzyl chain in the same positions was not intrinsically toxic. Therefore, the presence of an alkyl chain that increases the degree of lipophilicity can favour the cross and perhaps the injury of the outer membrane that represents the ideal target for new approaches [40].…”
