The macro flow planktometer: A new device for volume and fluorescence analysis of macro plankton including triggered video imaging in flow

Abstract: A newly developed macro flow planktometer is described for measuring plankton organisms with a size of 100-2,000 pm by the flow principle in timesaving and noncontact fashion. Based on the method of changes in electric resistance (Coulter principle), the device allows determination of the body volume of living organisms in their natural medium (fresh or seawater) at a measuring rate of up to 50 organisms per second. Simultaneous, laser-excited fluorescence measurement permits quantitative detection of fluoresc… Show more

“…A cocktail of beads of three different sizes (10,15, and 20 µm in diameter), which cover the size range of C6 glioma cells routinely used in our laboratory, was mixed to determine accuracy (i.e., coefficient of variation; CV), linearity, and reproducibility of the volume measurement. According to the specifications of the manufacturer, the CV of these beads is smaller than 2%.…”

“…When an image is flashed onto the target, the computer initiates its storage in the RAM memory and its display on the image monitor. A detailed description of the instrument is given in (Hüller et al, 1994). The instrument is mainly suited for analyzing and imaging particles and organisms in the range of 100µm to about 4000 µm diameter.…”

“…Manual microscopy has been the primary method for phytoplankton identification and enumeration 6 until the development of automated microscopy instruments, 7 and still remains the gold standard approach. 8 For accurate taxonomic identification in both manual and automated approaches, a trained taxonomist is required.…”
