The Method of Microbiological Soil Investigations within the Framework of the Project “Microbiome of Russia”

Abstract: The methods of Russian soil biology for the purposes of the investigations of soils microbiome were studied. The most important methods are cross-disciplinary investigations of plant and microbial interactions, the role of microbiome in the forming of soil fertility and carbon cycle. For the purposes of the most complete description of soil microbiome functions we suggest to combine the methods of metagenomics (for the purposes of the assessment of the phylogenic diversity of the microorganisms), biomarkers an… Show more

“…Cellulose-decomposing microorganisms -at Hutchinson's medium, the percentage content of soil foul particles reflecting the development of free-living nitrogen fixers of the Azotobacterecea family, at Ashby's nitrogen-free medium, the total number of bacteria was determined by the photocolorimetric method in P.A. Kozhevin modification using fluorescein diacetate [13][14][15][16]. The potential activity determination of nitrogen fixation was carried out by the acetylene reduction method, CO2 emission from the soil -by the absorption method [17] with the titrimetric end.…”

“…The dynamics of development and the number of individual types of microbes in greenhouse are determined by both abiotic and anthropogenic factors: tillage, number, and forms of plant residues of the precursor culture, as well as root exudates of vegetative species (Adams, 1990;Kostyuchenko and Lyakh, 2017;Rose et al, 2003). The influence of these factors can lead to changes in the dominant species, the parameters of the structure of soil biocenosis and the formation of specific functional rows of micromycetes, including toxigenic or pathogenic for cultivated plants (Estifanos et al, 2018;Chernov et al, 2017).…”
