“…Limited molecular studies of few Ca Ex-PAs and pleomorphic adenomas have shown relatively high abnormalities on the long arms of chromosomes 8 and 12, and the short arm of chromosome 17 (Mark et al, 1980;Johns et al, 1996;Yamamoto et al, 1996;El-Naggar et al, 1997;Gillenwater et al, 1997). We reasoned that detailed analysis of these chromosomal regions might provide further information on their involvement in the evolution and progression of Ca Ex-PA. Our objectives, therefore, were to determine the sequential genetic changes at certain regions of chromosome arms 8q, 12q, and 17p in PA and Ca Ex-PA and to correlate the results with various clinicopathologic factors of these tumors in the largest series investigated to date.…”

“…Fifteen microsatellite markers on chromosome arms 8q, 12q, and 17p were chosen based on recent information from molecular and cytogenetic studies of pleomorphic adenomas and Ca Ex-PAs (Mark et al, 1980;Bullerdiek et al, 1988Bullerdiek et al, , 1989Sreekantaiah and Sandberg, 1991;Sahlin et al, 1994;Johns et al, 1996;Yamamoto et al, 1996;El-Naggar et al, 1997;Gillenwater et al, 1997). Primers for polymerase chain reaction amplification of the microsatellite markers were obtained from Research Genetics (Huntsville, AL).…”

“…As first pointed out by Mark et al (1980), it frequently shows chromosomal abnormalities. Among the cases with abnor malities, rearrangements of chromosome 8 with a breakpoint at q 12 and rearrangements of chromosome 12 with a breakpoint at q 13 -*q 15 predominate (Mark et al, 1988), allowing the two main types of chromosomal rearrangements to be distinguished (Bullcr diek et al.…”

“…However, the situation closely resembles the cytogenetics of human pleo morphic adenomas (Mark et al" 1980(Mark et al" , 1988Bullerdiek et al" 1987). Although both tumors described here are not benign tumors, both neoplasms may have originated from benign tumors, and it is possible that canine mammary tumors, human pleomorphic adenomas, and some types of human breast can cers in their early stages share some common pathways of cyto genetic origin, in the sense that they might all originate from a cell with a single chromosome abnormality.…”

“…1987). Because about one half of all human salivary gland pleomorphic adenomas show specific chromosome aberrations (Mark et al, 1980(Mark et al, . 1988Bullerdiek et al, 1987), it is tempting to assume that canine mammary tumors display a similar pattern of cytogenetic aberrations.…”
