The MYC-YBX1 Circuit in Maintaining Stem-like Vincristine-Resistant Cells in Rhabdomyosarcoma

Abstract: Rhabdomyosarcoma (RMS) is a pediatric soft tissue sarcoma that causes significant devastation, with no effective therapy for relapsed disease. The mechanisms behind treatment failures are poorly understood. Our study showed that treatment of RMS cells with vincristine led to an increase in CD133-positive stem-like resistant cells. Single cell RNAseq analysis revealed that MYC and YBX1 were among the top-scoring transcription factors in CD133-high expressing cells. Targeting MYC and YBX1 using CRISPR/Cas9 reduc… Show more

“…26 Additionally, the interaction between YBX1 and MYC increased the stemness of rhabdomyosarcoma and led to vincristine resistance. 27 While the role of YBX1 in the progression of various human solid malignancies has been observed, to the best of our knowledge, this study provides the first clear evidence for the role of YBX1 in promoting stemness in ICC. Furthermore, the in vitro and in vivo experiments provide substantial evidence, and several ICC datasets provide these correlations, particularly demonstrating the correlation of YBX1 expression with stemness, the PI3K/AKT pathway and the WNT/β-catenin pathway in ICC cells at the cellular level.…”

“…This implies the existence of a potential reciprocal regulatory loop between Myc and YBX1, which could play a role in maintaining antiapoptotic signaling in tumors. In vincristineresistant rhabdomyosarcoma (RMS) cells, heightened CD133-positive stem-like cells revealed MYC and YBX1 as critical transcription factors [72]. Targeting the MYC-YBX1 axis reduced stem-like traits, suggesting YBX1's pivotal role and potential as a therapeutic target in RMS treatment.…”
