The novel gene fad104, containing a fibronectin type III domain, has a significant role in adipogenesis

Abstract: A novel gene named fad104 (factor for adipocyte differentiation-104), whose expression level quickly increased in the early stage of adipogenesis, was isolated and characterized. The deduced amino acid sequence of fad104 revealed the possible presence of a fibronectin type III domain and transmembrane domain. The expression of fad104 was detected in adipocyte differentiable 3T3-L1 cells but not observed in the non-adipogenic cell line NIH-3T3. Moreover, the ability of 3T3-L1 cells to differentiate declined wit… Show more

“…Three small interfering RNA (siRNA) sequences (adr 1-3) were designed to target regions of fndc3b (1, 627-645 bp; 2, 1,813-1,831 bp; 3, 3,391-4,009 bp ) as described 15 and were synthesized by GenePharma.…”

“…Reverse-transcription reactions were performed as described. 15 Realtime PCR was performed using a standard SYBR Green PCR kit protocol on a Rotor-Gene RG-3000A (Corbetter Research) thermocycler. The relative expression of miRNA compared with u6 was calculated using the 2 Ϫ⌬⌬Ct method.…”

“…104 The girl was found to carry a C-terminal truncated variant of the LRRC8A gene, and this truncation was identified as being the direct cause of the disease. Despite the role of the LRRC8 family in B cell maturation, the LRRC8 family has also been associated with adipocyte differentiation as well as lymphocyte and monocyte activation [105][106][107] In Lrrc8a -/-mice prenatal/postnatal mortality is increased and although the mice appear normal at birth and feed normally their growth became retarded, the hind legs turn weak, the stratum corneum is thickened, cysts are formed in the kidneys and the mice become sterile (no ovarian corpora lutea). 108 The LRRC8 family is composed of 4 transmembrane segments located at the N-terminal half of the protein and as illustrated for LRRC8A in Figure 11A a leucine rich repeat domain carrying up to 17 leucine-rich repeats (only 6 are shown in LRRC8A) located at the C-terminal end.…”

“…SNP rs11926347, in an intron of ABCC5 (OMIM 60521), and SNP rs6795506, near the 5' end of AHSG (OMIM 138680), had extremely small p-values (Table 8). Among the other nine genes, ADIPOQ (OMIM 605441, 612556) has been widely reported to be associated with obesity and diabetes [21-24]; FNDC3B (OMIM 611909) is involved in positive regulation of adipogenesis [25]; and DGKG (OMIM 601854) and AHSG (OMIM 138680) have been reported to be associated with obesity-related metabolic traits [26,27]. The remaining genes have no reported relation to obesity-related metabolic traits based on our literature review.…”

“…Some of the genes had been well-characterized in prior studies, e.g. FNDC3B, which is involved in adipogenesis [25]. However, the functions of most of the genes were not yet explicitly clear at the time of the analysis.…”
