The NS1 protein of H5N6 feline influenza virus inhibits feline beta interferon response by preventing NF-κB and IRF3 activation

Abstract: Despite the apparent lack of a feline influenza virus lineage, cats are susceptible to infection by influenza A viruses. Here, we characterized in vitro A/feline/Guangdong/1/2015, an H5N6 avian influenza virus recently isolated from cats. A/feline/Guangdong/1/2015 replicated to high titers and caused CPE in feline kidney cells. We determined that infection with A/feline/Guangdong/1/2015 did not activate the IFN-β promoter, but inhibited it by blocking the activation of NF-κB and IRF3. We also determined that t… Show more

“…The innate immune system recognizes invading viruses ( Hale et al, 2010 ; Iwasaki and Pillai, 2014 ) through multiple mechanisms. The non-structural NS1 inhibits type I IFN production ( Haye et al, 2009 ; Pekosz et al, 2012 ) by inactivating transcription factors ( MacEwan, 2002 ; Wang et al, 2017 ) such as IRF3 ( Wang et al, 2017 ), AP1, and NF-κB (miR-34) ( Taganov et al, 2006 ). Pattern recognition receptors (PRRs) recognize viral RNA ( Thompson et al, 2011 ; Okamoto et al, 2017 ) to activate a specialized immune response at mucosal surfaces to combat viral invasion.…”

“…The main ORFs of the virus were cloned and then ligated into the p3×Flag-CMV10 and pEGFP-C3 vectors. The feline interferon-associated luciferase reporter plasmids (IFN-β-Luc, NFκB-Luc, IRF3-Luc, and AP1-Luc) have been described in previous studies [ 41 , 42 ].…”

“…Besides the virus circulation in the poultry population by direct and indirect contacts between farms, LBMs all over China play the upmost important role in the disease transmission and as the sources of human infection. Particularly during the fourth and fifth epidemic waves, the Chinese authorities strengthened the National and Regional prevention and control measures by temporary closure and disinfection of LBMs, suspension of all live poultry trade, emergency monitoring of poultry and the culling of flocks in the case of HPAI A(H7N9) (Wang et al, 2017).…”
