The Nucleocapsid Region of HIV-1 Gag Cooperates with the PTAP and LYPXnL Late Domains to Recruit the Cellular Machinery Necessary for Viral Budding

Abstract: HIV-1 release is mediated through two motifs in the p6 region of Gag, PTAP and LYPXnL, which recruit cellular proteins Tsg101 and Alix, respectively. The Nucleocapsid region of Gag (NC), which binds the Bro1 domain of Alix, also plays an important role in HIV-1 release, but the underlying mechanism remains unclear. Here we show that the first 202 residues of the Bro1 domain (Broi) are sufficient to bind Gag. Broi interferes with HIV-1 release in an NC–dependent manner and arrests viral budding at the plasma me… Show more

“…2E and column 5 of Fig. 2G), indicating that the reported interaction between the NC domain of Gag and Bro1 domain of ALIX (45,46) was not sufficient to recruit detectable levels of ALIX in this system. The ALIX PRD contains a P(S/T)AP motif that is capable of binding to the TSG101 subunit of ESCRT-I (18), although the motif is not essential for ALIX function in HIV-1 budding (19,20).…”

“…61,68 ALIX and Tsg101 are two components necessary for the MVB biogenesis and for retroviral budding that associate with p6 for Tsg101 and for p6 in cooperation with NC for ALIX. 87,88 As mentioned before, the Bro1 domain of Alix interacts with the NC domain of HIV-1 Gag and in that manner cooperates to recruit the ESCRT via the p6 domain for retroviral budding. [87][88][89] Actin and actin-binding proteins are found inside HIV-1 virions 15,121 raising the question of a potential role of actin cytoskeleton dynamic in either Gag trafficking or in virion budding.…”

“…6C). Moreover, it has been shown by others that overexpression of full-length ALIX (57), as well as a fragment thereof (58), has a negative effect on the release of wt HIV-1. However, ALIX overexpression failed to increase the presentation of the SL epitope derived from GagSL-GFP wt (Fig.…”
