For sequence analysis of PAPP-A fragments generated by autocleavage, antigen was affinity-purified on anti-PAPP-A\proMPB [28] immobilized on CNBr-activated Sepharose (Pharmacia) according to the manufacturer's protocol. Bound protein was eluted with 200 mM glycine, pH 2.2, separated on 10-20 % reducing SDS\PAGE, blotted on to a PVDF membrane and stained with Coomassie Brilliant Blue.…”
