The RANKL expression and osteoclast in alveolar bone of rat diabetic model at different mechanical force application

Abstract: Background: Diabetes is a serious and important public health problem, especially in relation to dental treatment. Because of its complications in periodontal tissue, diabetes can be contraindicated in patients undergoing orthodontic treatment. The receptor activator of nuclear factor-κb ligand (RANKL) is an essential cytokine inducing osteoclastogenesis. Osteoblasts produce this cytokine which has been suggested to play an integral role in osteoclast activation during bone remodeling of orthodontic tooth move… Show more

“…In addition, experimental animals were induced with Streptozotocin (SC-200719, Santa Cruz) for 5 consecutive days at doses of 40, 35, 30, 25, and 20 mg/kg BW intraperitoneally followed by incubation for 7 days. 17 Diabetes in experimental animals was characterized by blood glucose levels >300 mg/dL.…”

“…A 30-g force of orthodontic appliance was applied in line with a previous study for 7 days. 17 In diabetes cacao groups, cacao extract was given orally using a gastric probe for 7 days after the orthodontic appliance was inserted. The doses of cacao extract were 125, 250, and 500 mg/kg BW.…”

“…The experimental animals were euthanised using ether anaesthesia; then the jaw was cut in the mandibular incisor region. The sample was prepared using 10% formalin for 24 hours, decalcified using 14% EDTA, for 30 days, dehydrated, cleared, and embedded in paraffin, then cut in the longitudinal direction with a thickness of 5 microns for the Hematoxylin-Eosin (HE) painting procedure [12]. Staining procedures were performed using HE to determine the histological feature of osteoclasts.…”
